dc.description.abstractPráce se zabývá studiem diagnostiky hereditárních nádorů prsu a vaječníku pomocí sekvenačních metod. Diagnostika hereditárních nádorů přechází v současnosti na sekvenování nové generace, které se stalo hlavní metodou při diagnostice dědičných predispozic pro vznik karcinomů prsu a vaječníku. Případný nález patogenní varianty je ověřován metodou přímé sekvenace. Praktická část se zaměřuje na návrh primerů a optimalizaci PCR, sekvenaci vybraných úseků genů a ověření patogenní varianty pomocí sekvenačních metod.cs
dc.description.abstract-translatedThe work deals with the study of the diagnosis of hereditary breast and ovarian cancer syndromes by means of the sequencing methods. Nowdays the diagnosis of hereditary breast and ovarian cancer syndromes makes for the next generation sequencing that has become the main method at the diagnosis of hereditary predispositions for a rise of breast and ovarian cancer syndromes. A potential finding of the pathogenic variant is verified by the direct sequencing method. The practical part is aimed at the proposal of primers and the optimalization of PCR, sequencing of selected parts of genes and the verification of the pathogenic variant by means of sequencing methods.en
