dc.description.abstractPráce se věnuje problematice velkých genových přestaveb a jejich detekci pomocí kvantita-tivní PCR. Byly analyzovány vzorky tří pacientů s podezřením na genetické aberace v konkrétních oblastech genomu. U všech vzorků byla detekována odchylka v relativní kon-centraci DNA ve srovnání s kontrolou, což potvrzuje přítomnost delecí nebo duplikace. Vý-sledky byly v souladu s předchozími nálezy, čímž se potvrdila vhodnost metody pro cílené potvrzení strukturálních změn. Práce současně poukazuje na možnost využití qPCR pro ori-entační stanovení rozsahu genové aberace.cs
dc.description.abstract-translatedThe paper deals with the problem of large gene rearrangements and their detection by quanti-tative PCR. Samples of three patients with suspected genetic aberrations in specific regions of the genome were analyzed. In all samples a deviation in relative DNA concentration com-pared to the control was detected, confirming the presence of deletions or duplications. The results were consistent with previous findings, confirming the suitability of the method for targeted confirmation of structural changes. At the same time, the work highlights the possi-bility of using qPCR for the orientation determination of the extent of gene aberration.en
