| dc.description.abstract | Cílem této práce bylo zjistit, zda je kolonková metoda se silikátovou membránou vhodná pro izolaci molekul mikroRNA z plazmy pacientů léčených pro melanom a zda poskytuje dostatečný výtěžek pro diagnostické a prognostické stanovení pacientů. Pro izolaci molekul miRNA z plazmy získané z plné krve byla použita souprava značky Qiagen a parametry izolované RNA (koncentrace a čistota) byly měřeny na přístroji Nanodrop 1000. Práce je rozdělena do části teoretické a praktické. V teoretické části je popsána patogeneze nádorového onemocnění a charakteristické znaky maligně transformovaných buněk. Dále je v této části kapitola zaměřující se na druhy rakoviny kůže, podrobněji pak na samotný melanom. Ke konci teoretické části je popsána miRNA a její vztah k rakovině, metody stanovení těchto molekul a koncept tekuté biopsie. Praktická část začíná definováním cílů a metodikou práce. Dále je podrobný popis izolace nukleových kyselin na silikátovém povrchu. Následuje analýza a vyhodnocení výsledků a statistické vyhodnocení naměřených hodnot. Bakalářská práce je zakončena diskusí metodických problémů a výsledků. | cs |
| dc.description.abstract-translated | The aim of this work was to determine whether the column method with silicate membrane is suitable for the isolation of microRNA molecules from the plasma of patients treated for melanoma and whether it provides sufficient yield for diagnostic and prognostic determination of patients. A Qiagen kit was used to isolate miRNA molecules from plasma obtained from whole blood, and the parameters of the isolated RNA (concentration and purity) were measured on a Nanodrop 1000 instrument. The bachelor thesis is divided into theoretical and practical part. The theoretical part describes the pathogenesis of cancer and the characteristics of malignantly transformed cells. In this part there is a chapter too focusing on the types of skin cancer, in more detail on the melanoma itself. At the end of the theoretical part is miRNA and its relation to cancer, the concept of liquid biopsy and methods for the determination of these molecules are described. The practical part begins with the definition of objectives and methodology of the work. The following is an analysis and evaluation of the results and a statistical evaluation of the measured values, which are part of the appendix. The bachelor thesis ends with a discussion of methodological problems and results. | en |
