dc.description.abstractTato práce se zabývá tématem volné cirkulující DNA (cfDNA) v laboratorní diagnostice tekutých biopsií s důrazem na nádorová onemocnění. Práce je rozdělena na teoretickou a praktickou část. Teoretická část této bakalářské práce se zaměřuje na systematický přehled aktuální literatury a výzkumných studií, které se především zabývají koncentrací cfDNA v časné diagnostice, sledování průběhu onemocnění a určování prognózy. V práci je také popsána struktura a funkce DNA, její vznik a uvolňování DNA do cirkulace. V rámci praktické části byly hodnoceny naměřené koncentrace cfDNA, izolované od pacientů s maligními lymfoproliferacemi a kvantifikované dvěma kvantifikačními metodami. Výsledky byly porovnány s klinickými daty pacientů, kteří byli léčeni na Hematologicko-onkologickém oddělení Fakultní nemocnice Plzeň. Byl prokázán vztah koncentrace cfDNA s klinickými parametry, jako je stádium onemocnění a odpověď na léčbu. Nebyla prokázána spojitost koncentrace cfDNA s věkem a pohlavím pacientů.cs
dc.description.abstract-translatedThis theses focuses on circulating free DNA (cfDNA) in the laboratory diagnosis of liquid biopsies with an emphasis on cancer diseases. The work is divided into a theore-tical and a practical part. The theoretical part focuses on a systematic overview of current literature and research studies, which mainly deal with the concentration of cfDNA in early diagnosis, monitoring and prognosis of the diseases. The thesis also describes the structure and function of DNA, its formation and DNA release into the circulation. As the practical part, the measured concentrations of cfDNA, isolated from patients with malignant lymphoproliferations, were evaluated using two quantification methods, and the results were compared with the patient's clinical data who were treated at the Haematology-Oncology Department of the University Hospital in Pilsen. The concentration of cfDNA has been shown to correlate with clinical parameters such as disease stage and response to treatment. The relationship of the cfDNA concentration with the patient's age and sex was not observed.en
